The default in /etc/cups/printers.conf (or classes.conf) is the
*server* default (also known as the network default printer in
CUPS 1.2).
The lpstat -d command reports the *user* default which may be
different. The "lpoptions -d printername" command sets the
user default printer, which is stored in /etc/cups/lpoptions when set
by root or ~/.lpoptions (CUPS 1.1.x) or ~/.cups/lpoptions (CUPS 1.2)
when set by an ordinary user.
