/etc/printcap is meaningless to configure CUPS.
It is only written by cupsd to provide backward-compatibility
for applications which read the print queues from /etc/printcap

I wonder why you think about how cupsd stores its files?
I guess you like to do some external special changes with
the print job files which can confuse a running cupsd and
result arbitrary problems in the printing system.
I recommend to use CUPS command line tools (or equivalent CUPS
library calls) for what you like to do with the printing system.
