If you mean, does CUPS work in a virtual machine, absolutely yes.  Just make sure that you configure the virtual machine with its own address (i.e. not a NAT configuration, but a bridged configuration).

We regularly test CUPS on multiple Intel-based operating systems using virtual machines...
