If you have configured one of the printers then open a web browser and visit the following URL : <http://127.0.0.1:631>. From the CUPS web interface you will be able to copy the connection URL for the printer. You will also have the option to delete the print queue. This will allow you to setup the other print queue using what ever method you are comfortable with. Finally, you will be able to retrieve the connection URL for this second printer from the CUPS web interface.

If you are having problems with the CUPS web interface then the following command in the terminal should list the connection URL's for any CUPS queues configured on your system :

	lpstat -v

In order to configure your printers with PrinterSetup you will require either the connection URL's for your printers or to have already setup the way you want in in cups so that PrinterSetup will be able to extract this information. One option is to setup one printer at a time and use PrinterSetup to extract the settings from CUPS. Then you could edit the automatically generated PSF's and alter the queue names.

I hope this helps.
