If you are developing a driver, start by reading the CUPS PPD
Extensions help file, available on your system at:

     http://localhost:631/help/spec-ppd.html

That said, there is 0 support for setting custom options from a GUI
on current MacOS X releases.  You can set them from the command-line,
but that is all...
