I told you to check all the listed directories, not only the last one!
Please also check the permissions from /opt/cups.

The CUPS scheduler run as root, not necessarily all filters and backends. Filters and backends normally run als lp user.

I assume one of your filters cannot access the cups spool directory. Therefor not only the spool directory permissions are deciding but also the parent directory permissions.
